Description

Doplet-3 is an injectable formulation of cholecalciferol (Vitamin D3) at 5 mg per mL, intended for intramuscular (IM) or subcutaneous (SC) use to rapidly correct vitamin D deficiency, especially when oral absorption is inadequate or in patients who need parenteral vitamin D supplementation. 


Ingredients

Uses / Indications

Doplet-3 injection is used for:

  • Treatment of vitamin D deficiency or insufficiency

  • Situations where oral vitamin D is poorly tolerated, absorption is impaired, or rapid correction is needed 


Dosage

  • The exact dose and schedule should be determined by a healthcare provider based on serum vitamin D levels, clinical status, and patient factors.

  • Common approach: a high dose injection (e.g. 5 mg = 200,000 IU equivalent) given as single or repeated injections under monitoring.

  • Maintenance oral vitamin D / supplementation often continues after injection.


In Case of Overdose

  • Excess vitamin D can lead to hypercalcemia: nausea, vomiting, weakness, frequent urination, dehydration, confusion, arrhythmias, kidney damage

  • Management: discontinue further supplementation, restrict calcium, provide IV hydration, possibly corticosteroids or bisphosphonates under physician direction


Missed Dose

  • Since this is often an infrequent injection, a “missed dose” protocol is less relevant

  • If missed, follow physician advice — do not double the next dose to compensate


How To Use / Administration Instructions

  1. Inspect ampoule: the solution should be clear, colorless, and free of particles

  2. Use aseptic technique to open and draw up the dose in a sterile syringe

  3. Administer via intramuscular (IM) injection (or SC if approved)

  4. Injection should be done by a qualified healthcare professional

  5. Avoid rapid injection to reduce discomfort


When Not to Use / Contraindications

  • Known hypersensitivity to cholecalciferol or formulation excipients

  • Hypercalcemia or hypercalciuria

  • Disorders with risk of vitamin D excess (e.g. sarcoidosis, certain granulomatous diseases)

  • Renal impairment with reduced excretory capacity (unless under specialist guidance)

Serious / rare:

Precautions & Warnings

  • Monitor serum calcium, phosphate, and renal function periodically

  • Use with caution in patients with renal disease, and avoid concomitant high-dose calcium without supervision

  • Avoid overlapping high-dose oral vitamin D or supplements without physician oversight

  • Pregnant/lactating women: only use if benefits justify risk and under medical supervision


Drug Interactions

  • Thiazide diuretics: may increase risk of hypercalcemia

  • Corticosteroids: may antagonize vitamin D absorption or effects

  • Certain anticonvulsants (e.g. phenytoin, phenobarbital): may accelerate vitamin D metabolism

  • Magnesium deficiency, proton pump inhibitors, etc.: may affect absorption/metabolism


Storage / Disposal

  • Store ampoules in original packaging, protected from light and heat, at recommended temperature

  • Do not freeze

  • Use ampoule immediately after opening; do not reuse

  • Dispose of used ampoule and needles in sharps / medical waste container
